A new form of bovine rhodopsin crystals with p22121 symmetry was obtained and for the first time not only images but also electron diffraction pattern could be obtained.

Out of a large number of images 20 crystalline areas were chosen, amplitudes and phases were extracted and a projection map to 5 Å was calculated. The most important conclusions are:

  • The molecules in this crystal form seem to be more densely packed leading to higher order of these crystals.
  • The quality of the extracted amplitudes and phases depends on careful boxing and unbending of the crystals.
  • The projection amplitudes were found to be reliable to 5 Å.
  • The phase error of unique Fourier components and the phase difference between two halves of data showed that phases were reliable to 5 Å.
  • In the new projection map helix 5 seems to be more highly tilted and between helices 3 and 5 a clear peak is present suggesting that helix 3 is prolonging the arc shaped feature towards helix 5.
  • Additional density is detected in the region of helix 6.
