Application to Antibody Engineering

Characterisation of bacteriophage-infectable mutD5 strains.
For use with phage-antibodies, bacteriophage-infectable mutD5 strains were required.


We constructed and characterised 2 strains:

KD1122pCJ : trpA, leu, arg, argE, his, thi, thr, spcA, mutD5, pCJ105 (F', ChlR)

NR9046mutD5: ara, thi, supE, rif, nalA, metB, argE(am), delta-prolac , F'prolac

Maintenace media (KD1122pCJ):

M9 salts, 1 mM Mg2SO4, 20 µM CaCl2, 5 µg/ml thiamine, 20 µg/ml chloramphenicol, 50 µg/ml spectinomycin, 1.6 mg/ml Cas amino acids (Sigma), 40 µg/ml L-tryptophan, 2% (w/v) glucose.

Mutation media:

2xTY, 20 µM thymidine
