RNA-protein Complex Structures
The first RNA-protein complex to be crystallised was a complex of E. coli glutaminyl-tRNA synthetase with its cognate tRNA (Rould et al.1989). Perone et al. (1988) overproduced tRNA(Gln) in E. coli, purified it to homogeneity and crystallised it with glutaminyl-tRNA synthetase. Around the same time the Strasbourg group determined the structure of the complex between yeast aspartyl-tRNA sysnthetase and its cognate tRNA (Ruff et al., 1991). The deterimation of these two structures was a major breakthrough in the field. Both structures show how the tRNA synthetase directly recognise the anticodon nucleotide. Subsequently in 1994 the structure of seryl-tRNA synthetase was determined in complex with its cognate tRNA (Bior et al., 1994). This enzyme recognises the congnate tRNA in a unique way. The extra loop of seryl-tRNA but not the anticodon loop is recognised by the enzyme.

In 1994 two more RNA-protein complex structures were reported. Valegaard et al. (1994) soaked an RNA hairpin representing the initiation site of assembly into a crystal of the MS2 bacteriophage capsid. The structure shows how the RNA hairpin binds to capsid protein around a dyad axis. We also reported the crystal structure of the spliceosome U1A protein in complex with a fragment of U1 snRNA (Oubridge et al., 1994). U1A contains an RNP domain or RRM (RNA recognition motif) which is one of the most commonly found RNA binding motif. Chemically synthesized RNA was used for both crytals. Crystal structures of more than twenty RNA-protein complexes have been reported to date. The atomic resolution structures of the 50S and 30S ribosomal subunits reprent a big step forward in our understanding of RNA-protein recognition and the archtecture of large RNA-protein assemblies
