A combination of direct-electron detection and statistical movie processing to compensate for beam-induced movement allows ribosome structures to be determined from cryo-EM data to resolutions that were previously only attainable by X-ray crystallography.

The Bayesian framework expresses the entire reconstruction workflow in one optimization target and learns the corresponding parameters from the data. Thereby, the need for user expertise is alleviated and arbitrariness is reduced. Results indicate that the Bayesian approach reduces overfitting, yields objective resolution estimates, and detects smaller classes in structurally heterogeneous data sets.
