Lab Resources
This page provides protocols and other resources that we hope will be of use to the scientific community.
Protocols
- Purification of ZZ tagged proteins from S.cerevisiae
- Protocol for purifying ZZ (protein ZZ) tagged dynein constructs. Derived from that reported in Reck-Peterson et al (2006).
- Microtubule gliding assay using GFP tagged kinesin or dynein
- GFP tagged motor proteins are attached to a glass coverslip via anti-GFP antibody. They drive the gliding of fluorescently labelled microtubules (see below for an example movie).
Educational materials
- Exploring DNA
- This is an interactive tutorial that introduces students to DNA. It was initially written for a museum in Cambridge, UK to celebrate the 50th Anniversary of the Watson and Crick's 1953 paper. More recently it was ported from Chime to Jmol by Frieda Reichsman and has been used in a number of college classes around the world.
Movies
Gliding assay: rhodamine labelled microtubules moved by gfp tagged dynein motors attached to the coverslip via an anti-GFP antibody. The brighter end of the microtubules are the plus ends. Click here to download movie
Single molecules of rhodamine labelled dynein (green) and cy5 labelled kinesin (red) moving along the same axoneme (bundle of microtubules). Note how the two motors move in opposite directions. Click here to download movie
